Cytokinins occur naturally in living plants, where they help regulate growth and cell division. One of them, kinetin (also known as furfuryladenine), drew interest in skincare as an ingredient for ageing skin. Pyratine LLC presents itself as the original developer of both kinetin and zeatin, another cytokinin, for therapeutic skincare [9]. That is the company's own account and has not been independently confirmed. The company's founding year remains unknown. The company frames its history as a progression. It points to its work on kinetin for ageing skin as the starting point, and describes PyratineXR as the step towards what it calls second-generation, improved cytokinins. The company also describes itself as a specialty therapeutic developer working on technologies that target unmet needs in dermatology. Much of that work, by the company's account, takes place in Europe rather than the United States. Pyratine says its scientific community is based in Olomouc, in the Czech Republic, where it runs a full research and development laboratory on site at the Institute of Experimental Botany [8]. Under the arrangement it describes, Pyratine funds the research done with the institute's scientists and in return receives the rights to all resulting discoveries. The company reports that this collaboration has produced more than a dozen new compounds screened for biological activity and for lack of toxicity to cells. Two of them, it says, are ready for commercial use: Pyratine itself and a second cytokinin known as 4HBAP. The company also names the Beijing Genomic Institute and the Greek Hellenic Research Society among its research partners. In February 2008 the Journal of Drugs in Dermatology published a clinical study of topical Pyratine 6 for improving the appearance of sun-damaged skin, by McCullough, Garcia and Reece [4]. The next generation was built on a different molecule, furfuryl tetrahydropyranyl adenine, which researchers studied under the code name PRK-124. A lotion containing 0.125 percent of the compound became the basis of PyratineXR. In May 2009 the same journal published a study by Ortiz, Elkeeb, Truitt, Hindiyeh, Tran and Weinstein on the lotion's effect on the signs and symptoms of rosacea. According to the German distributor, the cytokinin technology was examined in controlled clinical studies at medical schools, including the University of California, Irvine [4]. A longer trial followed. In June 2010 the journal published a 48-week study, led by Tremaine and colleagues, on the long-term efficacy and safety of the lotion in mild-to-moderate rosacea. As summarised in the company's announcement, subjects saw a mean 44 percent reduction in the severity of facial redness and a mean 89 percent reduction in the number of inflamed spots by week 48 [10]. The commercial launch came the following month. Pyratine's positioning rests on measurement. The company argues that many claims in skincare are made but few are substantiated, and says it does not rely only on subjective panels of consumers or dermatologists to judge its products. Instead, it says, it uses laboratory instruments to measure water loss from the skin and reductions in redness, and high-resolution photography and silicone profilometry, a technique that takes a silicone impression of the skin surface, to measure fine lines, wrinkles and roughness. It also states that PyratineXR contains a patented concentration of furfuryladenine, though no patent number is given [5]. According to the distributor, the active has been through a range of safety tests, including human aberration testing, the Ames mutagenicity test (a standard laboratory screen for substances that could cause genetic mutations) and eye sensitivity testing [5]. The distributor's summary of the study results lists 44 percent less visible redness, 89 percent fewer inflamed spots, 27 percent fewer visible small veins, 86 percent smoother skin, 41 percent more skin moisture and 22 percent fewer fine lines. The first two figures match the 48-week rosacea study; the distributor does not say which study each of the others comes from. Pyratine LLC goes further in its own claims. On its US site it calls PyratineXR a medically proven treatment for rosacea and the only known rosacea treatment that helps eliminate the condition's three key symptoms: facial redness, acne-like spots and spider veins [7]. It also says its two cytokinins, Pyratine and 4HBAP, have been clinically tested at leading universities and shown more benefits against the signs of ageing than earlier compounds, including Renova, which it describes as the only FDA-approved anti-ageing product [8]. The 2010 launch announcement named PyratineXR Moisturizing Lotion as the flagship of the new line.
